Linearity and Optimum-Sampling in Photon-Counting Digital Holographic Microscopy

چکیده

In the image plane configurations frequently used in digital holographic microscopy (DHM) systems, interference patterns are captured by a photo-sensitive array detector located at of an input object. The object information these is localized and thus extremely sensitive to phase errors caused nonlinear hologram recordings (grating profiles either square or saturated sinusoidal) inadequate sampling regarding coverage (undersampled around Nyquist frequency arbitrarily oversampled). Here, we propose solution for both recording problems through implementing photon-counting (PCD) mounted on motorized XY translation stage. such way, inherently linear (because wide dynamic range PCD) optimum sampled (due adjustable steps) holograms configuration recorded. Optimum estimated based numerical analysis. validity proposed approach confirmed experimentally.

Digital in-line holographic microscopy.

We first briefly review the state of the art of digital in-line holographic microscopy (DIHM) with numerical reconstruction and then discuss some technical issues, such as lateral and depth resolution, depth of field, twin image, four-dimensional tracking, and reconstruction algorithm. We then present a host of examples from microfluidics and biology of tracking the motion of spheres, algae, an...

Digital In-line Holographic Microscopy

Three-dimensional imaging with micron resolution and simultaneous tracking of many (hundreds) objects (bubbles, beads, algae, bacteria etc.) is now possiblein situ not only with a benchtop microscope, but also remotely in such environments as the deep ocean with a submersible version of the instrument. The instrument is very simple in its hardware requirements and an efficient software package ...

Speckle-field digital holographic microscopy.

The use of coherent light in conventional holographic phase microscopy (HPM) poses three major drawbacks: poor spatial resolution, weak depth sectioning, and fixed pattern noise due to unwanted diffraction. Here, we report a technique which can overcome these drawbacks, but maintains the advantage of phase microscopy - high contrast live cell imaging and 3D imaging. A speckle beam of a complex ...

Photon Counting Sampling of Phase Space

First complete experimental characterisation of the quantum state of a single light mode was demonstrated by Smithey et al. In their work, numerical tomographic algorithms were applied to reconstruct the Wigner function from homodyne statistics. Recently, a novel scheme for measuring the quantum state of light by photon counting has been proposed. In contrast to tomographic techniques, the newl...

Cell motility studies using digital holographic microscopy

Studies of cell movement have many applications, such as invasion of malignant cells into healthy tissue and leukocyte migration into inflammatory sites. Cultured adherent cells usually move over their substrate in a crawling motion that varies greatly between cultures due to factors such as cell type, malignancy, mutations, confluence, substrate, or external chemical or mechanical stimuli. Cel...
